Abstract
This experiment was conducted in Plant Tissue culture Laboratory / Horticulture and Landscape Department / Faculty of Agriculture /Kufa University during 2013-2014 .The aim was to study the effect of culture medium supplemented with NaCl salt, Methyl jasmonate and ascorbic acid on survival percentage, shoots number, its fresh and dry weights and its chlorophyll content of citrus rootstock Troyer citrange cultures under in vitro conditions . This experiment was carried as factorial using complete randomized Design with two factors, the first : NaCl at concentrations ( 0 , 75 or 150) mM and the second with combinations of (control , 2 mM of ascorbic acid ; 25 µM methyl jasmonate ; 50 µM methyl jasmonate ; 25 µM Methyl Jasmonate + 2 mM of ascorbic acid and 50 µM methyl jasmonate + 2 mM Ascorbic acid .) Results showed negative effects for NaCl on studied growth parameters of Troyer citrange rootstock cultures .It was found significant reduction in survival percentage, shoots number, its fresh and dry weights, and its chlorophyll content with an increase in NaCl levels in multiplication medium . They were the lowest values at 150 mM NaCl compared with the highest at control treatment ( 0 mM NaCl). Results also showed difference in responses of the plant growth regulators combinations in culture medium, treatment of 25 mM methyl jasmonate + 2 mM ascorbic acid increased significantly most studied parameters, compared with the free of plant growth regulators medium. Interaction between concentration of medium salinity and combinations of growth regulators showed significant differences in studied parameters. Cultures of the interaction treatment between methyl jasmonate and ascorbic acid (alone or together) with each concentration of salt were showed significant superiority on other treatments which gave an increase in survival percentage , shoots number , its fresh and dry weights, These results indicated the important role of methyl jassmonate and ascorbic acid in decreasing the adverse effects of NaCl salt.
