Effect of some Metal Ions Reducing and Chelating Agent on Collagenase Activity Extracted from Fish Wastes and Studying it’s Amino Acid Content | ||

| Abstract | ||
| The effect of metallic ions on collagenase activity were studied varied for both enzymes. Enzyme W kept full activity at concentration of 5 mM with the existence of potassium, magnesium and calcium ions. Cupric and aluminium ions showed inhibitory effect on the activity of enzyme W where residual activity reached 45.1 and 32.3 , 25.1 and 20.2% when enzyme was incubated with cupric chloride and aluminium chloride with concentrations of 1 and 5 mM, respectively. Chelating and reducing materials showed activating effect on enzyme, where the residual activity attained 101% when incubated with 5 mM 2-Mercaptoethanol. Enzyme also were maintained full activity when incubated with PMSF. As for isomer C, iron and zinc ions with concentrations of 1 and 5 mM showed obvious activating effect on enzyme where the residual activity ranged between 109.1-122.6%. The residual enzyme activity was observed to elevate when incubated with magnesium and calcium chlorides separately at concentrations of 1 and 5 mM where the activity fall between 109.1 and 125.1%. Chelating and reducing materials did not influence enzyme activity which maintained totally when incubated L-Cysteine, Urea and PMSF while EDTA played an inhibitory role on enzyme activity. Results revealed the variation in amino acid profiles for both enzymes W and C. An elevation was observed in percentages of Threonine, Serine, Glutamic acid, Aspartic acid, Leucine and Lysine in both enzymes. Other amino acids varied in percentages of existence in both enzymes. | ||
