Neuroprotective Effects of Hypericum triquetrifolium Turra Against Pentylenetetrazole-induced Seizures in Chicks
The present study was carried out to evaluate the safety and neuroprotective effects of aqueous extract of Hypericum triquetrifolium (HT) in chicks with pentylenetetrazole (PTZ) induced convulsive seizures. Sixty broiler chicks were randomly divided into six groups. The first negative control (NC) group received normal saline, while the second positive control (PC) group received PTZ (80 mg/kg). The third group was treated with carbamazepine (50 mg/kg) orally for six days. The fourth, fifth, and sixth groups were given HT extract at doses of 200, 400, and 800 mg/kg orally for six days, respectively. Two hours following last treatment dose, PTZ was injected to induce convulsive seizures in groups 3-6. Behavioural responses were monitored for thirty minutes, and mortality for three hours. The results showed that HT delayed the onset of convulsive seizures and improved the survival rate at 200 and 400 mg/kg (p < 0.05), and at 800 mg/kg (p < 0.001). GABA levels were elevated at 800 mg/kg (P < 0.01), while glutamate was reduced at 400 (P < 0.05) and 800 mg/kg (P < 0.01). MDA levels decreased at 400 (P < 0.01) and 800 mg/kg (P < 0.001), while TAO-C levels increased at 800 mg/kg (P < 0.05). IL-1β levels were reduced at 400 and 800 mg/kg (P < 0.05). In conclusion, HT extract appears relatively safe and exerts neuroprotective effects against PTZ-induced convulsions in chicks by modulating GABA neurotransmission, oxidative stress, and neuroinflammation; however, the present findings do not establish its superiority over carbamazepine.
