| Abstract | ||
| This study was conducted at Tissue culture Lab. –Date Palm research Center Basrah University to elucidate the effect of high temperature which used in sterilization by autoclaving on stability of main components in nutrient media in date palm tissue culture cv. Ashkar ,and consequently the effect on characteristics of embryogenic callus. In this study the main components of nutrient media which include vitamins (thiamine, biotin, pyridoxine and nicotinic acid), plant growth regulators (auxin NAA and Cytokinin 2ip) and other organic compounds (Adenine sulfate and mesoinositol) were sterilized by Millipore filters and by Autoclaving. Fresh and dry weight of embryogenic callus, number and length of somatic embryos were determined and compared to the results which obtained from using autoclave in sterilization. The results showed there was no significant difference in fresh and dry weight of embryogenic callus between the two sterilization methods. The medium which has adenine sulfate and mesoinositol sterilized by Millipore filters showed a superior significantly compared to other treatments in term of number of somatic embryos which was (33.67) embryos, while the media which has vitamins sterilized by Millipore filters gave a highest average of embryo's length which was (4.33) cm which differs significantly than other treatments. The results demonstrated that’s unfavorable to sterilize vitamins, adenine sulfate and mesoinositol by autoclaving, while its preferable to be sterilized by Millipore filter. The plant growth regulators NAA and 2ip showed a stability toward the high temperature. | ||
