Abstract
جمعت (50 ) عينة (ماء وتربة ) من مناطق مختلفة من العراق . أظهرت (5)عزلات قابليتها على النمو على وسط الأملاح المعدنية المضاف له الميثانول كمصدروحيد للكاربون والطاقه وبكثافة نمو مختلفة. أكدت الصفات المظهرية والفحوصات الكيموحيوية عائديتها للجنس Pseudomonas ( KK1-KK5 ) ، تمت دراسة المحتوى الوراثي للعزلات ، لمعرفة دور البلازميدات في اظهارصفة أ ستهلاك الميثانول ، وأظهرت نتائج الترحيل الكهربائي احتواء العزلات على حزمة بلازميدية واحده.اختيرت العزلة KK5)) وذلك لكفاءتها وقابليتها للنمو بكثافة على وسط الميثانول الانتقائي وبتركيز (0.8 % ) لأجراء تجارب الأقتران والتحييد. أ ظهرت نتائج الأقتران ا ن بلازميد العزلة KK5) ) له القدرة على التعبير المظهري في بكتريا ( MM294 E.coli ) أي ا نها ذات مدى مضيفي واسع ، وان الجينات الوراثية المسؤولة عن اظهار صفة ا ستهلاك الميثانول كانت بلازميدية الموقع ، وهذا ما اكدته نتائج تجارب التحييد .
