| Abstract The influence of the basal medium and different plant growth regulators on micropropagation of nodal explants from mature vine of Bougainville glabra cv. Sanderiana Variegata was investigated. Concentration of BA(0.5 ، 1.0 ، 1.5 ، 2.0 mg l−1) were used to optimize the proliferation phase. The number of shoots was dependent on the BA concentrations and best results were obtained with 1.5 mg l−1 BA. Shoot length was shorter with the higher BA concentrations, in both culture mediums (MS and WPM). Shoot elongation was best achieved when multiple shoots were cut into small pieces and sub cultured on elongation medium(MS) supplemented with 1.5 mg l−1 BA and 0.75 mg l−1 NAA. The transfer of in vitro shoots to rooting media, containing different concentrations of Naphthalene acetic acid (NAA) produced complete plantlets. The number of roots average and length were affected significantly by the NAA concentrations and was higher when 1.5 mg l−1 NAA was used only. | ||
