| This experiment was conducted during 2009 – 2010 in the tissue culture lab./ Dep. of plant production Technology / Technical college / Musayab. Vegetative buds of two potato cvs. (Dimant and Famosa) were sterilized with 5% sodium hypochlorite for 15 minutes. Sterilized buds were transferred to sucrose free MS medium supplemented with licorice extracts of different conc. ( 10, 20, 30, 40 and 50 ml / l ). Cultures incubated at temp. of 25ºC ± 2 with photoperiod of 16 h / day at 1000 lux light intensity for 60 day. On the other hand, merestematic apices ( 0.3 – 0.5 mm) were cultured on the same medium supplemented with 50 ml / l of licorice extract in a test tubes, incubated at the same conditions. The experiment comprises 2 trials, the first was the effect of licorice and cv. on the percentage of sprouted buds, leaves number, shoot number, nodes number/ plant, plant length (cm) and root length (cm). The second trial was to find out the effect of licorice conc.( 60, 70, 80, 90 and 100 ml / l ) on microcattings by the percentage of growing cuttings, leaves number, shoot number, nodes number/ plant, plant length (cm) and root length (cm). Results showed that the interaction of licorices conc. and cv. had a significant effect on all parameter. Licorice at 50 ml / l with Dimant resulted in the average of percentage of growing buds, leaves number, shoot number, nodes number per plant, plant length (cm) and root length (cm) reached ( 99.77%, 11.65, 2.88, 10.83, 12.89 cm and 3.69 cm) repectively. The interaction of 100 ml / l licorice with the cuttings taken from Dimant showed highest percentage of successful cuttings, leaves number, shoot number, nodes number/ cutting, plant length (cm) and root length (cm) that were ( 99.98%, 15.33, 2.71, 11.41, 13.11 cm and 6.71 cm) respectively. |
