| This experiment was carried out during year 0f 2009 – 2010 in the tissue culture lab./ Technical college / Musaib to study the ability micropropagation of Rosa damacena used single nodes at ( 0.5 – 1.0 cm) long. They sterilized by Ethyl alcohol ( 70% ) for 30 second with vibration, then washing with double distile water for 5 minutes, finaly soacking in sodium hypochlorite (1.5%) for 15 minutes. The nodes cultured on MS medium which contains NAA at ( 0.0, 0.5 or 1.0 mg /l ) with BA at ( 0.0, 0.5, 1.0 or 2.0 mg / l ). Cultures were incubated at 25ºC ± 2 and 16 h / day photoperiod with 1000 lux light intensity for 60 day. The plantlets produced from tissue culture had been cutted with micro cuttings with (0.5 cm ) long and cultured on MS medium of best concentration. The first experiment was to study the effect of different levels of NAA and BA on percentage of successful nodes, leaves number, shoot number, nodes number/ plant, plant length (cm) and root length (cm). The second experiment was to study the effect of different levels of NAA and BA on percentage of growing buds, leaves number, shoot number, nodes number/ plant, plant length (cm) and root length (cm), after 45 day from culture. Results showed that the addition of NAA and BA BA had a significant effect on properties studies. But the interaction among different levels of The concentration of 1.0 mg/ l of NAA with 2 mg / l of BA gave the highest percentage of successful cuttings, leaves number, shoot number, nodes number/ plant, plant length (cm) and root length (cm) they were ( 99.91%, 7.86, 2.73, 4.66, 7.41cm. and 2.68cm. ) repectively. Also the same interaction of 1.0 mg/ l of NAA with 2 mg / l of BA had super passed to gave the highest average of percentage of growing buds, leaves number, shoot number, nodes number/ plant, plant length (cm) and root length (cm) they were ( 99.97%, 6.61, 2.22, 3.88, 5.11cm. and 2.01cm.) respectively. |
