The present study was conducted in the experimental tissue culture lab. , Plant
Production Technology Dept. , Technical College / Musaib , during 2006 – 2007. The study included several experiments using tissue culture technique for the micropropagation of native garlic cultivar ( Allium sativum L.) , through induction of callus from root tip and the subsequent regeneration of whole plants as well The outstanding results emerging from this study are summarized as follow : -
1- Sterilization treatments using sodium hypoclorite solution ( NaOCl ) at ( 0.2 , 0.4 , 0.6 0.8 and 1.0 % ) for ( 5 , 10 , 15 , 20 and 25 minutes ) , showed that this aseptic in the concentration of 0.8 % for 20 minutes gave highest percentages of clean and free of contaminants cloves .
2- Callus induction trial using ( 0 , 0.5 , 1.0 , 1.5 and 2.0 ) mg / l 2,4-D and ( 0 , 5 , 10 and 15 ) mg / l kinetin showed that 1.5 mg / l 2,4-D with 5 mg / l kinetin significantly boosted Callus induction percent and callus fresh weight .
3- Treatment with ( 0 , 2.5 , 5.0 , 7.5 and 10.0 ) mg / l BA refered that the favourable conc. of BA for shoot regeneration was 5.0 mg / l which did not vary with the treatment where BA conc. was 2.5 mg / l for shoot proliferation from callus percentage, number and length of shoot per callus piece
4- Shoots that produced from callus were rooted on MS medium (full and half strength) supplimented with (0, 0.5 and 1.0 ) mg / l NAA . Auxin- free in full strength MS medium resulted in highest mean of root number and root length . plantlets on the other hand were acclimatized , and the Survival percentage resulted was75% |
