The Role of Ca2+ ATPase and Calcium Ion in Gentamicin Nephrotoxic effect in Rat Kidney Hemogenates | ||

| Abstract Two studies performed, one in vivo in which different increasing doses of gentamicin sulphate of (5, 10, 20, 40 & 0) mg/kg B.W. injected subcutaneously daily for five days to five groups of five rats each representing (T1, T2, T3, T4 & control) group respectively in which Ca2+ ATPase actively have been determined in their kidney hemogenates. The second study was in vitro, in which increasing concentrations of gentamicin (10-5, 10-4 & 10-3)M pre-incubated in normal rat kidney hemogenate to investigate the effect of gentamicin on Ca2+ ATPase activity in kidney cell. The results showed a sensitive inhibition of the enzyme activity in kidney hemogenate both in vivo and in vitro studies positively proportional with the increasing doses or concentrations of gentamicin sulphate. Inhibition of Ca2+ ATPase may cause reduction of calcium pump which take place in plasma and mitochondrial membranes, leading to accumulation of high concentration of Ca2+ ion inside the nephrotic cell with all consequences of the reported harmful effect of this accumulation on the normal function of the nephrotic cell that may contribute in gentamicin nephrotoxocity and other toxic side effect, like muscle and middle ear. | ||
