Abstract
تضمنت الدراسة الحالية معرفة تاثير مستخلص التمر (الدبس) والعسل كمصدر للكاربوهيدرات في استحداث ونمو كالس اوراق نبات الكزبرة(Coriandrum sativum L.). النامي على وسط(MS) المدعم بمنظمات النمو (BA,NAA) بتركيز 1.0 ملغم /لترلكل منهما . اذ ابدى مستخلص التمر( الدبس) والعسل القدرة على استحداث ونمو الكالس باعتبارها مصدر للطاقة والكاربون بدلا من السكروز الذي يستخدم عادة في الزراعة النسيجية للنباتات . فقد اظهرت النتائج بان اضافة الدبس بتركيز 2مل /100مل من الوسط الغذائي قد حفز استحداث الكالس بنسبة اعلى من السكروز بدلالة الوزن الطري اذ بلغ الوزن الطري 3.630 غم بعد مرور 130 يوم من الزراعة، كما لوحظ بان اضافة الدبس بتراكيز عالية (4,6) مل /100مل من الوسط الغذائي ادت الى عدم استحداث الكالس،وتشير النتائج ايضا بان اضافة العسل وبتراكيز متعددة حفز نمو الكالس بدرجة اقل اعتمادا على الوزن الطري مقارنة باضافة السكروز 30 غم الى الوسط الغذائي ويبدو ان اضافة 1مل/100مل من العسل حفز نمو الكاس بدرجة مقاربة بذلك الكالس النامي على وسط MSالحاوي على السكروز اذ بلغ الوزن الطري(2.216)غم ، في حين لوحظ انخفاض في نسبة الاستحداث بدلالة الوزن الطري عند استخدام تراكيز عالية من العسل (4,6) مل / 100مل اذ سجل (0.700 و0.500)غم على التوالي .كما اظهرت النتائج بان المحتوى الكاربوهيدراتي للكالس النامي على الوسط الغذائي الحاوي على تراكيز متباينة من العسل قد تفوق المحتوى الكاربوهيدراتي للكالس النامي على السكروز والدبس . اذ سجل اعلى محتوى كاربوهيدراتي (2.460)ملغم /غم للكالس النامي على العسل بتركيز 6 مل /100مل من الوسط الغذائي . The present study included the effect of date palm syrup (Dibs) and honey as the sole carbohydrates source on the initiation and growth of callus Coriandrum sativum L. leaves using Murashige and Skoog (MS) medium that contains 1.0mg /L of (BA,NAA) .the results cleared that date palm syrup and honey have capable of callus initiation and growth as carbon and energy source instead of sucrose usually used in most plant tissue culture .The result indicated that addition (Dibs) of 2ml/100ml to the nutrient medium induced the initiation of the callus with higher than that in the presence of sucrose, the fresh weight reaches to 3.630g after 130 day of culturing .whereas adding of high concentrations of dibs (4,6)ml/100ml could not stimulate of the callus. Initiation and results indicated also that adding different concentrations of honey would induce ,but to a very little proportion that grown as compared with the addition of (30)g of sucrose (adding 1ml/100ml of honey induce callus growth similarity to the callus growing in (MS) medium that contains sucrose (the fresh weight reaches to 2.216g).whereas adding high concentrations of honey does show a decrease in the induction of the callus (4,6)ml/100ml , the fresh weight reaches to (0.573-0.700 g).The study also showed that the carbohydrate content of the callus grown in nutrient medium which contains different concentrations of honey may surpass that content of the callus grown in both sucrose and dibis .such a content shows a highest proportion of carbohydrate (2.460)mg/g for the callus grown in honey of concentrations (6ml/100ml) of nutrition medium.
