Abstract
abstract: To asses genetic relationship between 21 cultivars of potato micropropagated in vitro, one of DNA markers which based on polymerase chain reaction was performed namely :RAPD (Randomly amplified polymorphic DNA ) .To achive this genomic DNA from 6-10 plantlets were isolated , the average yields obtained were in the range of 200-300 m3g and with apurity ranging between 1.6-1.8.DNA polymorphism were scored within amplified fragment (their numbers and molecular sizes) using agarose gel electrophoresis after staining with ethedium.Among. 72 primers tested 34 generated polymorphic patterns and selected for further RAPD analysis .Atotal number of 352 Bands were produced across all 21 potato cultivars .only 61 bands were monomorphic ,whereas the others were polymorphic representing high degree of polymorphisim reaching 82.6% of the total bands .Genetic distance among the 21 cultivars ranged from 0.13 between the cultivars Timate and Fresco (showing 87% similarity) to 0.352 between the Lola and Atlas (64% similarity).Cluster analysis by the unweighted pair group method of arithmetic means (UPGMA based dendrogram) revealed six main clusters. Mirakel and Ajeba cultivars represented the first major cluster. IPA, Tinor and Diamont represented the second, fourth and fifth cluster respectively. Cardinal and Lola formed the sixth major cluster.while the other cultivars represent the the third major cluster.The overall analysis of the results revealed that the genetic relationships between the cultivars was related to their morphological characters and sometimes with their origin.
