| The aim of this study is to analyzed gene expression for two genes mainly responsible on fatty acid desaturases production (FADs) are (FAD2.2 , FAD3) for 10 Olivecultivars (Oleaeuropaea L.) are (Khaderi , Qaysi , Manzenillo , Baashiqi , Arabqween , Nabali , Labeeb , Dahkan , Shami and Sorani) Two methods forgene expression analysis were used, , first method was called Semi – quantitative and second method was called relative – quantitative ,both used on leaves and fruits for trees were fruitful. used in relative method (Real time PCR and Actinegene as Housekeeping gene). The results revealed that. Semi – quantitative analysis for FADs genes in leaves for olive cultivars revealed that Iraqi cultivarsBaashiqi and Dahkan were the least cultivars expression for FADs genes , that was clear on bands for these varieties which were not found or weaker in most replication cDNA per programs with primers genes and special in 25 cycle PCR programs for semi – quantitative analysis . Other cultivars were with high intensity or medium specially Shami , Manzenillo and Soraniculivers which had clear bands in most Semi – quantitative PCR programs for two primer genes, and this refers to their high gene expression for FADs genes in leaves of olive cultivars .Semi – quantitative gene expression for fruit olive cultivars showed that Shamiolive cultivar was had indensity and clear bands to most(FAD2.2 andFAD3) genes and cycles except 25 cycle for (FAD2.2 ) gene bands were weak if compared with the others olive cultivars. So was Khadericultivars had indensity bands to most Semi – quantitative cycles to most genes, While Semi – quantitative gene expression for Iraqi olive cultivarBaashiqi was high indensity and clear bands to FAD2.2gene. Arabqween variety was high gene expression level for FAD2.2 gene about 17.5 fold , while Baashiqicultivars was in the second gene expression level for majority of genes studies and on the other hand that results showed decrease gene expression level for the other varieties for whole genes studies because this varieties may be intered in matury stage and ended Biocollection for fatty acid desaturated and oil. While high for FAD3 gene expression was in Manzenello fruitcultivars reached about 21.5 fold. |
