| Esculetin added at the beginning of culture of Concanavalin-A stimulated lymphocytes showed a dose-dependent cytotoxic effect. Complete cytotoxicity was seen at 62.5 μM esculetin treatment, and the lethal concentration fifty (Lc50) was 7.5 μM. As little as 0.25 μM esculetin showed 6.6% cytotoxicity compared to the control. With CTLL-16, an IL-2 dependent cytotoxic cell line, growth didn’t demonstrate any significant cytotoxicity in the presence of esculetin. Rather, this cell line showed 8% growth stimulation at 0.25 μM esculetin treatment. In the complete allogeneic primary lymphocyte culture (MLC) response, complete inhibition was achieved with 32 μM esculetin, whereas in the secondary MLC, complete inhibition required 64 μM esculetin. Fifty percent inhibition of primary and secondary MLCs were achieved with 12 and 20 μM esculetin respectively. In two-ways MLC, it was shown that esculetin inhibition at low doses was partly due to cytotoxicity and the inhibition was mainly due to cytotoxicity at high esculetin doses.
